How to do reticulocyte count?

Hello everyone, how are you all? Hope you all are doing well and i am too. Today i would like to teach on how to do reticulocyte count.

What are reticulocytes?

Reticulocytes are immature red blood cells (RBCs) that are produced in the bone marrow and released into the blood.

Principle

Reticulocytes are juvenile red cells containing remnants of ribosomes and ribonucleic acid. Ribosomes react with some vital dyes like brilliant cresyl blue or new methylene blue to form a blue granular filamentous precipitate in vitally stained unfixed preparations.
A precipitin tube is taken and 2 drops of blood and 1 drop of cresyl blue stain mixed in that tube and it is kept in incubation at 37o C for 30-45 mins. Next a smear made and air dried and kept under micrscope and seen with oil immersion at 1000x. Reticuloytes are identified by their deep blue reticulated appearance. Minimum of 1000 red cells are counted in which how many reticulocytes are there we have to count.

So in the above images i marked with arrows are the reticulocytes. So in this field we have to count total red blood cells. Then in that red blood cells how many reticulocytes are there. So in above image, there are total of around 100 red blood cells and reticulocytes which are labelled with arrows are 14 reticulocytes. Like this we have to count 1000 red blood cells and number of reticulocytes in it.
So for example we got around 140 oit of 1000 then we have to calculation by 140/1000 and multiplied 100 = 14. So reticulocyte count is 14. Which is high.
An elevated reticulocyte count suggests hemolysis.
