Cuando Castellano se convirtió en jefe, negoció una división de responsabilidades entre él y Dellacroce. Castellano tomó el control de los llamados "crímenes de cuello blanco" que incluían malversación de valores y otras grandes raquetas de dinero. Dellacroce conservó el control de las actividades tradicionales de Cosa Nostra. Para mantener el control sobre la facción Dellacroce, Castellano confió en el equipo dirigido por Anthony "Nino" Gaggi y Roy DeMeo. El equipo de DeMeo supuestamente cometió de 74 a 200 asesinatos durante los últimos años setenta y mediados de los años ochenta.
Durante su régimen, Castellano amplió enormemente la influencia de la familia en la industria de la construcción. Su nueva alianza con la banda irlandesa-americana Westside hizo millones de dólares para la familia en contratos de construcción.
