I wonder, why can't you just pin down a strand of DNA and use an electron microscope (or some other very powerful microscope) to measure its size and directly look at it?

I wonder, why can't you just pin down a strand of DNA and use an electron microscope (or some other very powerful microscope) to measure its size and directly look at it?
You certainly can do that, but electron microscopes are very expensive and it takes a lot of effort to prepare DNA for viewing under one. The protocols I wrote about are fairly low cost, rapid, and are commonly done before extensive analysis like with an EM.
